I think it was the Arthur C Clarke story (The Defenestration of Ermintrude Inch -- or something like that... I think that's the title) that made me first aware of the word...

I was studying for my SATs from a book that had an... interesting... list of words in the vocab section. 'Defenestrate' never actually showed up on any SATs that I took, but it's still a cool word!
