As for strip size: you don't actually need a scanner much bigger than A4. I scan my strips in two halves and glue them together in an appropriate art program. When you figure out how to do it it dosn't take very long to do the wall thing. Also when you've got you're colour paletes up and running it dosn't take that long to colour the whole strip... Okay, here's a tip: When you a large, flat surface like a wall or a floor it'll look really flat as one colour. This doesn't happen much in black and white but sticks out like a sore thumb when the strips is in colour! So even tho your not shading the rest of the comic, add some basic light effects ...

The less colours you use the smaller the PNG file size. Using 256 colours, PNGs kick GIF files asses! Of course it's sometimes better to use JPEGs, but it is a VERY lossy (I don't think that's a real word) format and often shows around the text in a strip. Well I can't add much to what's been said except is best to have a number of art/editing apps. I have PhotoShop, Paint Shop Pro and Fireworks. PhotoShop is my main workhorse as it's FAR superior to the other two BUT Fireworks can convert PhotoShop PSD's to PNGs well (converting layers and everything...
